Centrifugal microfluidic-based 3D spheroid culture for rapid and high throughput generation
In this paper, we first present a centrifugal microfluidic-based spheroid (CMS) formation method for rapid and high throughput 3-dimensional (3D) spheroid cell generation. Circularly arrayed microwells were designed to allow even distribution of cells introduced at the center of a rotating platform and to provide identical hypergravity conditions at each well by centrifugal forces generated. Compared to the conventional well-plate (WP) spheroid formation method, CMS formation method significantly promotes cell aggregation, compactness, and size homogeneity with high production yields. We were able to generate concentric, janus, and mixed type of multicellular spheroids, in addition to single cell type ones, using human adipose-derived stem cells (hASCs) and human lung fibroblasts (MRC-5) by successfully creating cell-cell interfaces between the two cells, demonstrating versatility of CMS formation method.
