Evaluation of weaving nerve guide conduit using PCL porous yarn for sciatic nerve regeneration

In this study, we evaluated nerve guide conduit (NGC) which was fabricated using polycaprolactone (PCL) porous yarn via weaving method for nerve regeneration in rat’s sciatic nerve injury model with 10-mm nerve defect. The PCL NGCs was simply prepared by roll up the weaving sheet using stainless rod having 3-mm diameter. Characteristic of prepared NGCs were analyzed by permeability using fluorescence conjugated dextran, scanning electron microscope (SEM), in-vitro test cell penetration test using the weaving sheet, and in-vivo test using rat’s sciatic nerve injury model. The NGC was implanted in sciatic nerve injury model and sacrifice at 1, 6, 12 weeks respectively. Regenerated nerve was evaluate by electromyogram, H&E stain and toluidine blue stain after the NGT removal at rat’s sciatic nerve.
