혈액 뇌 장벽 모사를 위한 3차원 공배양 모델

The blood brain barrier (BBB) protects brain tissue from harmful materials by obstructing molecular transport and helps to keep homeostasis. The barrier property is mediated by the interaction of multiple cell types including brain endothelial cells, astrocyte and pericyte as well as the physiological environment of human brain. The animal model has been used for drug testing but they have the big problem of its genetic heterogeneity. Therefore, in vitro BBB model has been consistently necessitated for identifying their physiological roles in pathogenesis and applying to the drug screening platforms. Although several in vitro BBB models have been developed, replicating the native BBB properties still remains challenging. Here, we presented a three dimensional BBB model comprising of brain endothelial cell and astrocyte or pericyte with dynamic fluidic environment. We fabricated in vitro brain blood vessel by culturing human brain microvascular endothelial cells (hBMEC) in the luminar surface of collagen tubular channel. By co-culturing astrocyte or pericyte in the bulk collagen, we could demonstrated the three dimensional interaction of multiple cell types. Moreover, the endothelial cells were exposed to pulsatile shear stress similar to the physiological blood flow in the body. Our BBB model is believed to be a robust and reliable platform for screening the effect of neurological drugs in the future.
