PCR 열순환기 마이크로채널에서의 최적온도를 위한 수치해석에 관한 연구

A continuous flow polymer chain reaction(PCR) platform device with integrated heaters for DNA amplification has been designed and fabricated with the 3D printing technologies. The finite element method (FEM) was employed to analyze the temperature distribution in the DNA chip. Three temperature regions, which correspond to denaturation, annealing and extension, respectively, were formed in a stretch of microchannel by three independent heaters on which microfluidic device was fixed. The temperature of the PCR solution was periodically switched between 55 °C and 95 °C. The temperature distribution was measured with IR camera. The microchannel platform has a width of 300 μm, a depth of 300 μm, and a total length of 2.7 m with 27cycles. The flow rates for the experimental are 5 uL/min and 10 uL/min. We have successfully developed the PCR thermal cycler with manufactured the 3D printer.
